A study has been made of the effect of udder infections on the proportionate yield and composition of the milk of individual quarters of over 100 cows, and the data from eleven of these cows have been presented.

Typical examples have been given showing that subclinical Streptococcus and haemolytic Staphylococcus infections may persist for months and either cause a gradual reduction in the proportionate yield, and solids-not-fat percentage of a quarter or have no measurable effect, and that both types of reaction may occur simultaneously in the same udder.

Examples also show that following penicillin treatment of clinical cases caused by streptococci and staphylococci there is little recovery in the proportionate yield of the treated quarter until the following lactation, when recovery may be complete.

Lactic acid bacteria (LAB) can be isolated from different sources such as milk and cheese, and the lipolytic, proteolytic and glycolytic enzymes of LAB are important in cheese preservation and in flavour production. Moreover, LAB produce several antimicrobial compounds which make these bacteria interesting for food biopreservation. These characteristics stimulate the search of new strains with technological potential. From 156 milk and cheese samples from cow, buffalo and goat, 815 isolates were obtained on selective agars for LAB. Pure cultures were evaluated for antimicrobial activities by agar antagonism tests and for proteolytic activity on milk proteins by cultivation on agar plates. The most proteolytic isolates were also tested by cultivation in skim milk followed by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) analysis of the fermented milk. Among the 815 tested isolates, three of them identified as Streptococcus uberis (strains FT86, FT126 and FT190) were bacteriocin producers, whereas four other ones identified as Weissella confusa FT424, W. hellenica FT476, Leuconostoc citreum FT671 and Lactobacillus plantarum FT723 showed high antifungal activity in preliminary assays. Complementary analyses showed that the most antifungal strain was L. plantarum FT723 that inhibited Penicillium expansum in modified MRS agar (De Man, Rogosa, Sharpe, without acetate) and fermented milk model, however no inhibition was observed against Yarrowia lipolytica. The proteolytic capacities of three highly proteolytic isolates identified as Enterococcus faecalis (strains FT132 and FT522) and Lactobacillus paracasei FT700 were confirmed by SDS–PAGE, as visualized by the digestion of caseins and whey proteins (β-lactoglobulin and α-lactalbumin). These results suggest potential applications of these isolates or their activities (proteolytic activity or production of antimicrobials) in dairy foods production.

The aim of this research was to enhance the nutritional quality of ewe milk fat by increasing potentially healthy fatty acids (FA) through diet supplementation with unprotected oil rich in linoleic acid, and without detrimental effects on animal performance. Twenty-four ewes were assigned to two high concentrate diets, control or supplemented with 6% sunflower oil (SO), for 4 weeks. No differences between treatments were found in milk production and dry matter intake. Although the SO diet increased milk fat percentage and tended to reduce milk protein concentration, it did not affect milk fat, protein or total solid yield. Most of the modifications in milk FA composition were addressed toward a potentially healthier profile: a decrease in C12:0 to C16:0 and a remarkable increase in the contents of cis-9 trans-11 C18:2 (from 0·94 to 3·60 g/100 g total FA) and trans-11 C18:1 (from 2·23 to 8·61 g/100 g total FA). Furthermore, the levels reached were maintained throughout the period monitored. However, the SO diet increased other trans C18:1 isomer percentages, too. The lack of differences between treatments in the in vitro ruminal fermentation parameters, studied with batch cultures of rumen microorganisms, would indicate no negative effects on ruminal fermentation.

Chemical and physical changes that occur in Na caseinate (at 1 or 2% in water, pH 7·0) on heating in the range 120–150 °C were investigated by polyacrylamide gel electrophoresis, ion-exchange and gel-permeation chromatography, light scattering, u.v. spectroscopy, amino acid analysis, and the formation of pH 4·6 and 12% TCA-soluble N and 12% TCA-soluble P. The electropherograms of heated samples were smeared and indistinct suggesting intermolecular aggregation which was not reversed by 6 M-urea or SDS and indicating covalent bond formation; αs2-casein was especially sensitive. Aggregation was confirmed by ion-exchange chromatography and light scattering. Fragmentation of the caseins also occurred on heating, as indicated by the formation of pH 4·6 and 12% TCA-soluble N and by gel filtration. Formation of soluble N and dephosphorylation followed first-order kinetics. Concentrations of available lysine and available methionine were reduced by 10–15% on heating at 140 °C for 30min; chemical assays for arginine and tryptophan indicated increases, suggesting interference. Ultraviolet spectroscopy indicated a slight apparent increase in aromatic residues after heating at 140 °C for up to 60 min.

A survey was undertaken to update and extend available information on the vitamin content of pasteurized milk as produced at processing dairies in mainland UK and to investigate regional, seasonal and breed effects. The concentration of total retinol in milk from non-Channel Island (NCI) breeds averaged 61·9 βg/100g in summer and 41·2 βg/100g in winter. Concentrations of β-carotene were 31·5 and 10·5 βg/100g in summer and winter respectively. Concentrations of retinol in milk from Channel Island (CI) breeds were similar, but concentrations of β-carotene were on average 3 times higher. The concentration of vitamin D3 in milk from NCI breeds was 0·033 βg/100g in summer and 0·026 βg/100g in winter. There was no marked seasonal variation in the mean concentration of total vitamin C (14·5 βg/ml). Values for the concentration of B vitamins (βg/ml) were: folic acid 0·060, vitamin B12 0·0042, riboflavin 1·78, nicotinic acid 0·71, pantothenic acid 3·60, biotin 0·020, thiamin 0·46 and vitamin B6 0·61. Seasonal variation in the concentration was most marked for folic acid (c.v. 17·4%) and to a lesser extent for vitamin B12 (c.v. 10·3%). The only breed differences in the B vitamin content were for riboflavin and folic acid, the mean values obtained for milk from CI breeds being respectively 20 and 10 % higher than those from NCI breeds.

An examination was made of the effect of varying the protein, lactose and total serum ion content of milk on the rate of production of heat-induced acidity (as measured by pH decrease), on heat stability (as measured by coagulation time), on the relationship between coagulation time and initial pH of milk (as measured by coagulation time-pH curves) and on the interrelationship of these parameters. In addition, the effect on these parameters and their interrelationship of varying the composition and volume of the headspace atmosphere in contact with the milk was investigated. Explanations are proposed for the observed effects on heat stability of varying the composition of milk and the heating conditions, with special reference to the influence of heat-induced acidity.

Four multiparous Holstein cows, each equipped with a duodenal cannula and an ultrasonic mammary blood flow probe, were assigned to a 4×4 Latin square to measure the effects of duodenal infusions of Leu (0, 40, 80 and 120 g/d) on lactational responses and mammary metabolism of nutrients. Cows were fed a diet of 67·0% corn silage, 5·3% grass hay, 14·2% peas, 7·8% maize starch, 1·0% each of molasses, urea, and sodium bicarbonate and 2·4% minerals and vitamins. Requirements of the remaining 9 essential amino acids were met by infusing into the duodenum 323 g/d of a mixture of amino acids including, Ile, Val, Met, Lys, Trp, Phe, His, Thr, Arg, Tyr and Glu. Milk protein content and yield were highest with 40 g/d of Leu and then progressively declined. Milk fat content and yield were significantly decreased over the entire range of Leu infusions. Arterial concentrations of Leu increased linearly and corresponding mammary arterio-venous differences increased to a plateau with 80 g/d of Leu. As Leu infusions increased, extraction rates of Leu decreased linearly, whereas those of Ile, Val, Lys, Arg, Thr and Tyr were significantly increased. Leu was taken up by the mammary gland according to milk output with 0 and 40 g/d of Leu infused and then largely in excess. Based on responses of protein yield and mammary uptake to milk output ratio, Leu concentration in total amino acids absorbed in the small intestine needs to be close to 8·9% for optimal milk protein synthesis.

A sensitive, specific radioimmunoassay for bovine α-lactalbumin (α-la) suitable for measurement of serum, tissue culture media and milk was developed. α-La was not detected in serum from prepubertal male or female cattle, but was detected as early as 60 d of gestation in nulliparous Holstein heifers, the level being greatly increased during the last one third of gestation. Serum from cross bred beef heifers contained less α-la and it was not detected until late gestation. Concentrations of α-la in serum samples from pregnant multiparous Holstein cows decreased at drying-off and subsequently increased just before parturition. Secretion of α-la by mammary tissue explants from steroid-primed prepubertal Holstein heifers was induced by the addition of bovine prolactin, ovine prolactin or human growth hormone to tissue culture media.

A method is described for the quantitative determination of phosphatase in raw milk. Using this method, it has been found that phosphatase is sufficiently thermolabile to be destroyed completely by pasteurisation if this process is properly carried out. The absence of phosphatase from a sample of milk or cream indicates with a fair degree of certainty that the milk or cream has been heated sufficiently to destroy such pathogenic organisms as were originally present, though it is of course no guarantee that the product is free from these organisms at the time of testing.

A simple, qualitative test-tube method is described which may be used for differentiating between raw and heated milk, or between raw and heated cream, and which with slight modifications may also be used for distinguishing between butters made from raw or from heated cream.

Zn binding by whole bovine and human casein and by purified bovine caseins and whey proteins was investigated by equilibrium dialysis. Bovine αs1 casein had the greatest Zn-binding capacity (˜ 11 atoms Zn/mol). Protein aggregation was observed as Zn concentration was increased and- the protein precipitated at a free Zn concentration of 1·7 mM. Zn binding increased with increasing pH in the range 5·4–7·0 and decreased with increasing ionic strength. Competition between Zn and Ca was observed for binding to αs1-casein indicating common binding sites for these two metals. Bovine β-casein bound up to 8 atoms Zn/ mol and precipitated at a free Zn concentration of ˜ 2·5 mM, while K-casein bound 1–2 atoms Zn/mol. Whole bovine and human casein bound 5–8 atoms Zn/mol and precipitated at a free Zn concentration of ˜ 2·0 mM. Scatchard plots for Zn binding to caseins showed upward convexity, possibly due to Zn-induced association of caseins. Apparent average association constants (K¯app) for all caseins were similar (log K¯app 3·0–3·2). Enzymic dephosphorylation of αs1- or whole bovine casein markedly reduced, but did not eliminate, Zn binding. Thus, phosphoserine residues appeared to be the primary Zn-binding sites in caseins. With the exception of bovine serum albumin. which bound over 8 atoms Zn/mol, the bovine whey proteins, β-lactoglobulin, α-lactalbumin and lactotransferrin, had little capacity for Zn binding.

Listeria monocytogenes survived and, under most conditions, multiplied when inoculated directly into the cheese milk of laboratory made Camembert cheeses. The rate and extent of growth was reduced at lower storage temperatures. Significantly higher rates of growth occurred at the surface compared with the centre of the cheeses, and these were probably associated with increased pH and proteolysis at the cheese surface due to the mould ripening process. Similar results were obtained with Camembert cheeses surface inoculated after manufacture. There was also temperature-dependent growth of List, monocytogenes on a range of inoculated commercially manufactured soft cheeses. Significant growth occurred in Cambazola, French and English Brie, blue and white Lymeswold, French Camembert and Brie with garlic. Little if any growth occurred in blue and white Stilton, Mycella, Chaume and full fat soft cheese with garlic and herbs at the temperatures examined.

Whole milk from cows was concentrated 2-fold by reverse osmosis in a small pilot scale apparatus. All the main constituents of the milk were retained in the concentrate.

The experiment showed that protein was a major cause of blocking of the membrane. Laboratory trials were therefore made on the ultrafiltration of milk to remove the protein with the aim of developing a 2-stage process for the concentration of milk by ultrafiltration followed by reverse osmosis with subsequent recombination of the concentrates from the 2 operations. The rates of both processes and the retentions of the constituents of milk by the membranes were measured.

The use of a silica-based ion-exchange medium for the recovery of protein from wheys prepared by treatment with rennet or with acid has been investigated. The protein capacity of the medium whilst maintaining an adsorption greater than 75% during the passage of rennet-and acid-whey was 0·079 and 0·053 g/g respectively. Bovine serum albumin (BSA), α-lactalbumin (α-la) and β-lactoglobulin (β-lg) adsorbed from acid-whey were mostly recovered in yields greater than 93% in an undenatured form essentially free of lactose and milk salts by elution using 0·1 M-HC1. With rennet-whey, BSA and part of the α-la were not recovered in the native form, probably because of proteolysis during elution by enzymes originating in the rennet used in the manufacture of the whey. There was, however, no effect on the recovery of native β-lg adsorbed by the medium. An unidentified proteinaceous fraction, thought to be of casein origin, was also adsorbed and recovered from rennet-whey. Fractions of the adsorbed proteins could not be obtained by eluting with a gradient increase in NaCl concentration. Nevertheless, purified β-lg was obtained by passing a large quantity of whey through the medium. This protein had a high affinity for the functional groups of the medium, to the extent that other proteins initially adsorbed were subsequently displaced. Unidentified proteinaceous material in rennet-whey also had a high affinity for the functional groups of the ion exchanger but most of this fraction could be removed by a pretreatment procedure which involved passing the whey through the medium at pH 5·0; unidentified material was selectively adsorbed at this pH. The medium could also be used to produce a fraction containing a mixture of BSA and α-la but it was not possible to separate these proteins.

During a 12-month longitudinal study, bulk-tank milk was collected from organic (n=17) and conventional (n=19) dairy farms in the UK. Milk samples were analysed for vitamin A (retinol), vitamin E (α-tocopherol) and β-carotene content. The farming system type, herd production level and nutritional factors affecting the milk fat vitamin content were investigated by use of mixed model analyses. Conventionally produced milk fat had a higher mean content of vitamin A than organically produced milk fat, although there were no significant differences in the vitamin E or β-carotene contents between the two types of milk fat. Apart from farming system, other key factors that affected milk fat vitamin content were season, herd yield and concentrate feeding level. Milk vitamin content increased in the summer months and in association with increased concentrate feeding, whilst higher-yielding herds had a lower milk vitamin E and β-carotene content. Thus, conventional dairy farms in the UK produced milk with a higher vitamin A content, possibly owing to increased vitamin A supplementation in concentrate feeds. However, knowledge of the effects of season, access to fresh grazing or specific silage types and herd production level may also be used by all producers and processors to enhance the vitamin content in milk.

The concentration ranges in milk and cheese of a number of metallic elements relevant to the health of the consumer are dependent upon animal species, feeding, time of year, environmental conditions and manufacturing processes. We have measured the concentrations of selected trace elements – Al, Ba, Cd, Co, Cr, Cu, Fe, Mg, Mn, Ni, Pb, Pt, Sr and Zn – in raw ewes' milk and typical ewes' milk cheeses. Samples of milk, curd, whey, cheese after moulding, cheese after salting, commercial cheese (Pecorino), Ricotta, scotta and brine were analysed for seven complete cycles of cheese production. Samples of curd, whey, brine and waste product (i.e. scotta) were collected and analysed in addition to the cheese and milk to assess the distribution of each element in the main product, by-products and waste products during cheesemaking. Quantitative determinations were performed mainly by inductively coupled plasma atomic emission spectrometry. There were considerable differences among the trace element contents of raw ewes' milk and related products, indicating that manufacturing processes play a key role in the distribution of minor and trace elements. Two different approaches were used to assess the dietary intake of these elements associated with the consumption of Pecorino and Ricotta, and whether this had any nutritional or toxicological implications. The results indicated no toxicological risk and showed that, on the contrary, Pecorino may be a good source of several essential elements.

The effects of daily supplements of 300 g native and 2 hydrogenated cod-liver oils (CLO) to the diets of lactating cows have been examined. Native CLO supplements increased the proportion of propionate and decreased the proportion of acetate in the rumen and also decreased the yields in milk of fatty acids synthesized within the mammary gland and those derived from plasma triglycerides. These effects were reduced or eliminated when the hydrogenated oils were given instead of the native CLO. With all 3 supplements about 15% of the dietary C20and C22 acids was secreted in milk.

Both native and hydrogenated CLO supplements increased the concentrations of the cholesteryl ester and phospholipid components of the α-lipoproteins in the blood plasma. These components contained most of the C20 and C22 acids of the CLO supplements that were incorporated into the blood plasma lipids. With the native CLO supplement about half of the C20 and C22 acids present in the blood plasma were polyunsaturated.

There were no differences in the effectiveness of high-density plasma lipoproteins as activators of triglyceride emulsions for lipoprotein lipase hydrolysis whether the lipoproteins were from cows receiving the native or the hydrogenated CLO supplements or the control diets. Significant rates of hydrolysis by lipoprotein lipase could not be demonstrated using as substrates low- or high-density plasma lipoproteins or activated emulsions of their extracted lipids.

Additional in vitro experiments using activated triglyceride emulsions as substrates for mammary lipoprotein lipase showed that replacement of a soybean oil emulsion by an emulsion of native CLO reduced the rate of hydrolysis considerably, whereas an emulsion of hydrogenated CLO was without effect unless it was present at levels in excess of 50%.

It is concluded that the change in rumen fermentation induced by the polyunsaturated fatty acids of CLO is implicated in the decreased intramammary synthesis of fatty acids, but that additional information is required before the reason for the reduced mammary uptake of plasma fatty acids can be established.

Cows with subclinical intramammary infections were identified by milk bacteriology. The mastitis pathogens included Staphylococcus aureus (n=9), Streptococcus uberis (n=10) and coagulase-negative staphylococci (n=10). Samples of first fore milk, main flow milk and strippings milk fractions were collected from each quarter and laboratory measurements were made of electrical conductivity, milk fat concentration and somatic cell count. Conductivity measurements were corrected for milk fat concentration and within-cow inter-quarter conductivity ratios calculated. Repeatability estimates of all measurements between days were calculated. In the case of infected quarters, all conductivity values decreased markedly (P<0·05) from first fore milk to main flow milk fractions. Conductivity differences between quarters of infected cows were substantially lower during the main milk flow phase. For quarters infected with Staph. aureus an increase in conductivity was observed (P<0·05) from main flow to strippings fractions. For uninfected quarters, conductivity declined as milk fat concentration increased with successive milk fractions. Variation, both within and between milk fractions, was greater for somatic cell count than for conductivity. Differences in conductivity between milk fractions from individual infected quarters were not accounted for by changes in fat concentration and may result from the mixing of milk from infected and uninfected regions of the gland. Localized infection may produce a decrease in conductivity between fore milk and mid-flow fractions while differential drainage from an infection site in the secretory tissue may additionally produce an increase in conductivity from mid-flow to strippings fractions. Such changes may thus provide information on the location and magnitude of an infection. The results clearly demonstrate the importance of the milk fraction when using conductivity as a diagnostic of intramammary infection, the highest diagnostic sensitivity being achieved by using first fore milk samples.

Interest in selective dry cow therapy (SDCT) has been increasing owing to concerns over development of antimicrobial resistance. Implementation of SDCT, however, requires a quick and cost-effective on-farm method for identifying cows for treatment and cows that can be left without treatment. The objective of the present study was to evaluate the use of clinical mastitis (CM) history and somatic cell counts (SCC) from monthly Dairy Herd Improvement (DHI) records in identification of infected and uninfected cows at dry-off. A total of 647 Holstein cows were classified as uninfected or infected at dry-off based on CM history and varying number of monthly SCC records (with three different SCC cut-offs). Cows were considered uninfected based on the following criteria: (1) SCC <100 000 cells/ml and no CM during the lactation; (2) SCC <200 000 cells/ml and no CM during the lactation; (3) as criterion two, but additionally a cow was also considered uninfected if it experienced a case of CM during the first 3 months of the lactation and the SCC was <100 000 cells/ml for the rest of the lactation; (4) SCC <300 000 cells/ml and no CM during the lactation; otherwise they were considered infected. Infected and uninfected cows at dry-off were most efficiently identified using three months' SCC records with a threshold of 200 000 cells/ml for cows without CM during the lactation and a threshold of 100 000 cells/ml during the rest of lactation for cows with CM during the first 90 days in milk. Moreover, this criterion also most efficiently identified cows infected with major pathogens only at dry-off. The success of the criteria used for identifying infected and uninfected cows will, however, depend on herd characteristics, such as prevalence of infection and type of pathogens present in the herd.
