  • Prostaglandin E2 (PGE2) may play a major role in embryo development and the establishment of pregnancy in cattle. The biosynthesis of PGE2 implies the sequential transformation of arachidonic acid to PGH2 by cyclooxygenases (COXs), then the conversion of PGH2 to PGE2 by prostaglandin E synthases (PGESs). Quantitative RT-PCR was used to examine the expression of COX-1, COX-2, microsomal PGES-1 (mPGES-1), microsomal PGES-2 (mPGES-2) and cytosolic PGES (cPGES) mRNAs in day 7 in vitro-produced (IVP) embryos from oocytes collected by ovum pick-up in Holstein heifers. Transcripts for COX-2 and mPGES-1 were detected in all embryos, whereas transcripts for COX-1 and mPGES-2 were not detected and cPGESs were at the limit of detection in 40% of embryos. Levels of COX-2 and mPGES-1 mRNAs were significantly higher in blastocysts and expanded blastocysts than in morulae and early blastocysts. Furthermore, excellent-quality embryos (grade 1) displayed higher levels of both COX-2 and mPGES-1 than did embryos of good and medium qualities (grades 2–3). Our results suggest that bovine IVP embryos at the morula and blastocyst stages use exclusively the COX-2/mPGES-1 pathway for PGE2 biosynthesis, and that PGE2 is potentially involved in blastocyst expansion and developmental competence.

  • Quality of transgenic rabbit embryos with different EGFP gene constructs

  • The aim of this study was to compare the quality of rabbit transgenic embryos obtained upon microinjection of gene constructs containing different promoters and green fluorescent proteins (CMVIE–EGFP, PGK–EGFP and CMVIE–hrGFP). Developmental rate, total cell number in hatching blastocyst stage, number of apoptotic cells, diameter of embryos, transgene integration and transgenic mosaicism were investigated.

    The rate of rabbit embryos microinjected with the different gene constructs developed up to morula stage was significantly lower (p < 0.05) than that of intact (non-microinjected) rabbit embryos (66–74vs. 98%). The highest efficiency of transgene integration (15%) was found when the CMVIE–EGFP (DrdI) gene construct was used, however a significantly higher transgenic mosaicism (60%) was found in rabbit embryos using this gene. The lowest cell number was counted in rabbit transgenic embryos with CMVIE–rhGFP linearized by ScaI (115.0 ± 8.20), the highest cell number (134.0 ± 35.00) was detected in rabbit transgenic embryos carrying PGK–EGFP (Not I) gene. The highest number of apoptotic cells (2.6 ± 0.33) was recorded in rabbit transgenic embryos with the integrated CMVIE–EGFP (ClaI) transgene.

    Based on these results a more suitable gene marker for rabbit transgenic embryos production and selection is the CMVIE–EGFP (ClaI) gene construct. Prior to using microinjected embryos (for embryo transfer, vitrification or ESC isolation) it is necessary to pre-select microinjected embryos with evident transgenic mosaicism.

  • The objective of current study was to compare the nuclear configurations of canine oocytes recovered from between follicles after isolation. Follicles isolated were classified into follicle-S (follicles located in the ovarian surface) and follicle-I (follicles located inside the ovary) based on the follicle location in the ovary. Nuclear stages of canine oocytes recovered from follicle-S and follicle-I were examined by phase-contrast microscopy after isolation. Results demonstrated that canine GV stage oocytes can be classified into three types based on the status of the nuclear envelope, nucleolus, and chromatin: type A, type B, and type C. In follicle-S group, the majority (95.5%) of canine GV stage oocytes was of type B. All canine GV stage oocytes recovered from follicle-S (including type B and type C) were characterized by nuclear envelope disappearance prior to nucleolus collapse. In contrast, in follicle-I group, the majority (60.2%) of canine GV stage oocytes was of type C. Unexpectedly, a small proportion of canine GV stage oocytes from follicle-I (donated type A) were characterized by nuclear envelope disappearance following nucleolus collapse. In conclusion, nuclear configurations of each type of canine GV stage oocytes may differ from each other. Distributions of each type of canine GV stage oocytes may associate with the follicle location in the ovary.
