The antidotes dichlormid, MON-4660, CGA-154281, R-29148, and MON-13900 were tested in the greenhouse to protect Zea mays L. (corn) against RPA 201772 injury. High rates of RPA 201772 injured four Z. mays hybrids > 30%. R-29148 and MON-13900 were the most effective of the five antidotes evaluated. R-29148 applied at rates ⩾ 45 g ha−1 provided excellent protection against RPA 201772 injury and also prevented injury to Z. mays from diketonitrile, the active metabolite of RPA 201772. In laboratory studies, R-29148 did not alter absorption of 14C-RPA 201772 from soil; however, R-29148 significantly enhanced the rate of RPA 201772 metabolism and inactivation in Z. mays. The mixed function oxidase inhibitor piperonyl butoxide (PBO) increased RPA 201772 injury on all hybrids. These results demonstrate that Z. mays tolerance to RPA 201772 can be enhanced with the use of antidotes such as R-29148 and MON-13900, that R-29148 protects Z. mays from RPA 201772 and diketonitrile by the enhancement of metabolism, and that oxidative reactions may be involved in the metabolism of RPA 201772 in Z. mays.

Greenhouse and laboratory studies were conducted to determine the effects of bentazon on the crop safety, efficacy, foliar absorption, and translocation of thifensulfuron when it is applied to soybean, velvetleaf, and common lambsquarters. A metabolism study was conducted on soybean. Thifensulfuron applied at 1.1 g ai ha−-1 with 28% urea ammonium nitrate (2.5% v/v) and BAS-0904805 (Dash) adjuvant (0.63% v/v) reduced the growth of velvetleaf and common lambsquarters by an average of 91 and 84%, respectively. The addition of 420 g ai bentazon ha−-1 had no effect on thifensulfuron efficacy in the weed species. Soybean dry weights were decreased by 58% when thifensulfuron was applied at 2.2 g ha−-1 but were decreased by only 36% when bentazon, at 420 g ha−-1, was added. In the absorption study, the addition of bentazon reduced foliar absorption of 14C-thifensulfuron into velvetleaf and common lambsquarters 8 and 24 h after treatment (HAT), but absorption into soybean was not affected. Bentazon reduced the translocation of 14C from thifensulfuron out of the treated leaves of velvetleaf and common lambsquarters by at least 16 and 11%, respectively, beyond 24 HAT. Soybean translocated 18, 29, and 26% of absorbed 14C out of the treated leaflets 24, 72, and 168 HAT, respectively. These translocation values were reduced to 7, 12, and 11%, respectively, when bentazon was tank-mixed with thifensulfuron. Soybean metabolized 35% of recovered 14C-thifensulfuron by 24 HAT. Addition of bentazon did not change this level of metabolism. These studies suggest that the physiological basis for the decrease of soybean injury from thifensulfuron, when it is tank-mixed with bentazon, is decreased thifensulfuron translocation.

Growth chamber experiments were conducted to determine the effects of UV light and riboflavin on photolysis of 2,4-D applied to Zea mays leaves. Droplets of 100 mg L−114C-2,4-D were applied to Z. mays leaves with and without 10 mg L−13H-riboflavin and exposed to either UV-enhanced or UV-attenuated polychromatic light in a time-course assay. Photolysis of nonabsorbed 14C-2,4-D residues on Z. mays leaves was sensitized by riboflavin regardless of UV light regime, but a larger percentage of nonabsorbed herbicide was degraded under UV-enhanced light compared to UV-attenuated light. Riboflavin was almost completely photolyzed during the first 10 h of exposure; yet, photolysis of 14C-2,4-D surface residues in treatments containing riboflavin increased from 59% at 10 h of exposure to 87% at 42 h of exposure. In corresponding treatments without riboflavin, photolysis of 14C-2,4-D surface residues was 37% at 10 h of exposure and 84% at 42 h of exposure. In contrast, only 7% of the 14C-2,4-D deposited on glass microscope slides was degraded after 42 h of exposure in the absence of riboflavin, whereas 59% was degraded in the presence of riboflavin. Photolysis of 2,4-D on Z. mays leaves in treatments without riboflavin suggests that certain epicuticular component(s) of Z. mays acted as photosensitizers or catalytic agents that promoted photolysis of nonabsorbed 2,4-D residues.

Variability in weed control following pyrithiobac applications has been observed under field conditions. The influence of temperature on this variability was investigated. Results from field studies performed over two growing seasons identified plant and air temperatures at the time of herbicide treatment that correlated with whole-plant efficacy differences. Based on the field data, weed control with pyrithiobac was acceptable at application temperatures of 20 to 34 C. To investigate a potential source of thermal limitations on pyrithiobac efficacy, the thermal dependence of in vitro inhibition of acetolactate synthase (ALS), the site of action for pyrithiobac, was examined. A crude leaf extract of ALS was obtained from Amaranthus palmeri. Relative inhibitor potency (I50) values were obtained at saturating substrate conditions for temperatures from 10 to 50 C. Regression analysis of field activity against I50 values showed the two data sets to be highly correlated (R 2 = 0.88). The thermal dependence of enzyme/herbicide interactions may provide another means of understanding environmental factors limiting herbicidal efficacy and predicting herbicide inhibition at the whole-plant level.

Laboratory experiments were conducted to assess the influence of surfactants applied with or without nitrogen on MON 37500 foliar absorption by Bromus tectorum, Bromus japonicus, Aegilops cylindrica, Triticum aestivum, Chorispora tenella, and Lactuca serriola. MON 37500 absorption in B. tectorum and B. japonicus increased from 40% 24 h after treatment (HAT) to 48% 48 HAT, averaged across surfactants with no added nitrogen. Averaged across nitrogen source and species, nonionic surfactant, ethylated seed oil, and organosilicate provided comparable enhancement of MON 37500 absorption (56 to 68%), whereas crop oil concentrate provided only 27 to 29% absorption under the same conditions. Averaged across species and surfactant class, urea ammonium nitrate had the greatest effect on MON 37500 absorption (68%), compared to ammonium sulfate (59%) or no nitrogen (40%). Nitrogen, regardless of the type, significantly improved foliar absorption of MON 37500. MON 37500 absorption by species was 71, 63, 57, 57, 49, and 38% in C. tenella, B. japonicus, T. aestivum, A. cylindrica, B. tectorum, and L. serriola, respectively, when averaged across surfactants and nitrogen. Densely pubescent B. japonicus leaves did not retain significant amounts of MON 37500 following a primary leaf wash.

A kochia biotype from McDonough County, Illinois, was suspected to be resistant to both triazine and acetolactate synthase (ALS)-inhibiting herbicides. We performed greenhouse and laboratory experiments to confirm, quantify, and determine the molecular basis of multiple herbicide resistance in this biotype. Whole-plant phytotoxicity assays confirmed that the biotype was resistant to triazine (atrazine), imidazolinone (imazethapyr), and sulfonylurea (thifensulfuron and chlorsulfuron) herbicides. Relative to a susceptible kochia biotype, resistance to these herbicides ranged from 500- to > 28,000-fold. The kochia biotype from McDonough County also displayed high levels of resistance (2,000- to 9,000-fold) to ALS-inhibiting herbicides in in vivo ALS enzyme assays, indicating that resistance to these herbicides was site-of-action mediated. Results from chlorophyll fluorescence assays indicated that triazine resistance was also site-of-action mediated. Foliar applications of atrazine had little or no effect on photosynthesis in the resistant biotype, even when atrazine concentrations were 108-fold higher than needed to inhibit photosynthesis in the susceptible biotype. A region of the gene encoding the D1 protein of photosystem II and all of the open reading frame of the gene encoding ALS were sequenced and compared between the resistant and susceptible biotypes. Resistance to triazine and ALS-inhibiting herbicides in the kochia biotype from McDonough County was conferred by, respectively, a glycine for serine substitution at residue 264 of the D1 protein and a leucine for tryptophan substitution at residue 570 of ALS.

We conducted several experiments on linuron-resistant and -susceptible Portulaca oleracea and on atrazine-resistant and -susceptible Chenopodium album to determine their immediate and long-term responses to photosynthesis-inhibiting herbicides. Several photosynthesis-inhibiting herbicides were used, and O2 evolution was measured with a Clark-type O2 electrode. Resistance ratios (RRs) for P. oleracea, based on O2 evolution inhibition, were 8 and > 6 for linuron and diuron, respectively; > 800 for atrazine; and > 20 for terbacil. Linuron-resistant P. oleracea was negatively cross-resistant to bentazon and pyridate (RR = 0.5 and 0.75, respectively). Time-course measurements of fresh weight, photosynthetic CO2 assimilation, and photochemical efficiency indicated that linuron and atrazine inhibited electron transport in susceptible (S) P. oleracea and C. album, ultimately resulting in death. Measurements of photochemical efficiency and CO2 assimilation of linuron-resistant P. oleracea treated with linuron indicated a transient injury from which plants recovered within 14 d. Recovery of linuron-resistant P. oleracea from atrazine injury was more rapid than from linuron injury for all measured variables. Atrazine-resistant C. album had no cross-resistance to linuron. Sequence analysis of the D1 protein revealed that linuron-resistant P. oleracea had a serine-to-threonine substitution at position 264.

The ability to predict time of weed seedling emergence relative to the crop is an important component of a mechanistic model describing weed and crop competition. In this paper, we hypothesized that the process of germination could be described by the interaction of temperature and water potential and that the rate of seedling shoot and radicle elongation vary as a function of temperature. To test these hypotheses, incubator studies were conducted using seeds and seedlings of common lambsquarters. Probit analysis was used to account for variation in cardinal temperatures and base water potentials and to develop parameters for a new mathematical model that describes seed germination and shoot and radicle elongation in terms of hydrothermal time and temperature, respectively. This hydrothermal time model describes the phenology of seed germination using a single curve, generated from the relationship of temperature and water potential.

In glasshouse experiments, Panicum repens rhizomes with more than two nodes emerged from soil depths of 1 to 20 cm. Emergence from one-node rhizomes was greatly reduced at depths more than 5 cm. In contrast, culm emergence from ginger like rhizomes decreased when burial depth (BD) was greater than 20 to 30 cm. P. repens emerged from deeper soil levels with an increasing number of nodes per cutting. Culms emerged from 36% of total underground regenerative organs. About 75 and 95% of the emerged culms were recorded 30 and 60 d after burial, respectively, and 91% emergence was recorded from 20-cm BD. In field studies, only 9% of nodes produced culms following cross-plowing. About 61 and 67% of the emerged culms were recorded 50 and 60 d after land preparation, respectively. Results indicate that mechanical and chemical control methods could be most effective for P. repens when applied approximately 50 d after land preparation. Burying deeper than 30 cm by deep plowing and reducing rhizome length by cross-plowing might effectively reduce emergence of P. repens.

Competitive cereal cultivars are less susceptible than others to weed interference. Their characterization may provide selection criteria that can be used as guidelines to develop new, even more competitive cultivars. Root exudates are a potential means by which competitive cultivars reduce weed growth. The objectives of this study were to evaluate the effect of cereal root exudates on Brassica kaber (DC.) L. C. Wheeler growth, to isolate and characterize the allelochemical compounds released by spring cereal cultivars, and to determine if a relation exists between these allelochemicals and cultivar competitiveness. Highly competitive (HC) and lesser competitive (LC) cultivars of four crop kinds (Triticum aestivum L. [wheat], Avena sativa L. [wild oat], two- and six-rowed Hordeum vulgare L. [barley]) were selected based on previous work. Exudates from undisturbed root systems of B. kaber and cereals were collected and used in a bioassay test with B. kaber. Root exudates were analyzed for 16 common phenolic compounds using high-performance liquid chromatography (HPLC). Bioassays indicated that cereal exudates had no negative effect on B. kaber germination, but all concentrations of cereal root exudates inhibited B. kaber root and hypocotyl growth. As cereal root exudate concentration increased, B. kaber growth decreased. For each crop kind, B. kaber growth inhibition was greater with HC cultivars than with LC cultivars. The root exudates of all crop kinds and cultivars contained benzoic, caffeic, ferulic, o-coumaric, and vanillic acids as well as scopoletin. Para-hydroxybenzoic acid was found in exudates from T. aestivum, A. sativa, and two-rowed H. vulgare cultivars. Para-coumaric acid was not identified in root exudates from LC H. vulgare cultivars. Gentisic acid was produced by A. sativa and H. vulgare. Vanillic and o-coumaric acids along with scopoletin may be responsible for the allelopathic effects of H. vulgare, T. aestivum, and A. sativa cultivars. These three compounds may be useful as possible indicators of allelopathic potential of genotypes under development and thus considered for use in breeding programs.

Reports of unacceptable shattercane (Sorghum bicolor) control with acetolactate synthase (ALS)-inhibiting herbicides prompted the investigation of 29 fields in central and south-central Nebraska for ALS-resistant (ALSr) shattercane. These fields were located in three distinct geographical areas designated C, G, and P. Shattercane from 13 fields spanning all three areas was resistant to 80 g ai ha−1 (2 X field rate) primisulfuron. Accessions C and G were more resistant than accession P to primisulfuron and nicosulfuron. Accessions C and G were susceptible to imazethapyr, whereas accession P was resistant. The ALS resistance was associated with alterations in the ALS enzyme. Primisulfuron I50 values for ALS from ROX (forage sorghum), C, G, and P were 7, 8,510, 8,870, and 714 nM, respectively; nicosulfuron I50 values were 647, 4,110, 4,070, and 1,460 nM, respectively; and imazethapyr I50 values were 5,440, 13,100, 11,800, and 51,700 nM, respectively. Based on cross-resistance and enzyme sensitivities, at least two biotypes are represented in the three accessions of ALSr shattercane. Shattercane individuals from accessions C, G, and P were intercrossed to determine if the ALSr genes in each of the accessions were at independent loci. All the F2 populations were resistant to 80 g ai ha−1 primisulfuron, suggesting that the ALSr alleles in the three accessions are at the same locus or possibly linked loci. When the C, G, and P accessions were crossed with the wild type (WT), comparisons between the F1, susceptible, and resistant populations showed that primisulfuron resistance was expressed as a dominant, partially dominant, and additive trait for the C, G, and P accessions, respectively. The differences in ALSr allelic interactions indicate that primisulfuron resistance developed independently in each of the three accessions.

Detailed knowledge about the environmental conditions required for weed seed germination and establishment in soil is an important prerequisite for the development of integrated and biological weed control strategies. Germination and establishment of Amaranthus retroflexus were studied at different temperatures, planting depths, soil types, nitrogen supply, and water potentials. The minimum temperature for seed germination was > 5 C; maximum germination occurred between 35 and 40 C. At temperatures between 25 and 35 C, an additive effect on germination rate was observed when temperature and water availability were increased. For all soils tested, the percentage emergence of seeds placed on the soil surface and 4 cm deep was significantly lower than seeds placed between 0.5 and 3 cm. Emergence in the four sandy soils was generally greater than in the two heavier soils included in the study. There was a highly significant interaction between seed depth and soil type. Plant growth was also greatest in the lighter soils. Plant height, number of leaves, leaf area, fresh and dry weight, and nitrogen and carbon percentage in plant tissues of A. retroflexus increased significantly with increasing soil nitrogen supply.

The mode of action of BAS 517 in a susceptible plant species, corn, was investigated using an excised root system and 14C-tracer techniques. The root system of a tolerant species, soybean, was used for comparison. When UL-14C- glucose was used as a precursor, 14C incorporation into lipids was reduced in BAS 517-treated corn roots, although 14C incorporation from UL-14C-glucose into lipids was relatively low. Inhibition of 14C incorporation into water-soluble compounds was not definite because of a high degree of variability. Using 14C-acetate as a precursor, 49, 43, and 34% of the recovered radioactivity was found in the lipid fractions of root tips treated with 0, 1.0, and 10 μM BAS 517, respectively. In nontreated soybean root tips, 47% of the recovered radioactivity was found in the lipid fraction compared to 49% in root tips treated with 10 μM BAS 517. Further analysis of lipids showed that BAS 517 inhibited the incorporation of 14C from 14C-acetate into phosphatidylethanolamine, a phospholipid, whereas the labeling of sterols in treated corn roots was not adversely affected. Acetyl CoA carboxylase extracted from root systems of corn and soybean showed different sensitivity to BAS 517, suggesting its role as the herbicide target site and as a basis for the selectivity.

Emergence patterns of foxtail in spring wheat following soybean were evaluated for three seeding dates and three tillage regimes. Cumulative foxtail emergence, as a percentage of total plants emerged in the growing season, was generally not influenced by tillage regime throughout most of the emergence period, but when differences occurred, emergence was lower with no-till than with moldboard plow. Foxtail seedling densities were greater in no-till and chisel plow than in moldboard plow. Weed biomass and wheat yields were not affected by tillage regime. Delaying wheat seeding reduced foxtail percent emergence and emerged seedling density. Differences in emergence patterns of foxtail were attributable to thermal accumulation after seeding. Wheat yield was not influenced by seeding date in 2 of 3 yr.

Transgenic Nicotiana tabacum with tolerance to 2,4-D has previously been produced using a bacterial 2,4-D-dioxygenase gene (tfdA) driven by the 35S promoter of cauliflower mosaic virus. Using promoters from the Pisum sativum plastocyanin gene (petE) and an Arabidopsis thaliana histone gene (H4A), we demonstrate that similar protection from 2,4-D can be obtained in transgenic N. tabacum by targeting expression of tfdA to either meristematic tissues or chloroplast-containing tissues. As with the 35S promoter constructs, the plants are tolerant but not completely resistant; very young seedlings in particular are only slightly protected. However, the levels of tolerance observed could offer a useful degree of protection from accidental spray drift.

Kyllinga species are becoming more prevalent in turfgrass sites throughout North America. The effects of nitrate (50, 200, and 400 mg L−1), temperature (33/24, 25/17, 19/11 C day/night, respectively), and light on seed germination of three Kyllinga species were investigated in growth chambers. Nitrate concentrations did not stimulate Kyllinga species seed germination compared with untreated seeds. All Kyllinga species seeds failed to germinate in darkness but resumed germination once they were placed in light. This is an important pest management strategy because a dense, uniform turfgrass stand with its minimum light penetration to the soil would minimize Kyllinga species seed germination. Higher temperatures increased seed germination rate and percentage of each species after 8 wk. Maximal (> 90%) K. brevifolia germination occurred 2 to 4 wk after initiation in every seed study, whereas K. squamulata seeds germinated continuously. Minimal (< 10%) K. pumila seeds germinated until alternating diurnal temperatures were imposed.

A field study was established in southern Iowa in 1994 to study seasonal and long-term weed population dynamics on land being brought back into production after 8 yr as part of the conservation reserve program (CRP). The study was a split-plot design with four replications; two tillage regimes, two crop rotations, and three herbicide application methods were used. Even though the tillage regime did not influence individual weed population density throughout the study, the no-till (NT) regime had more weeds compared to conventional tillage (CT). However, when weeds were grouped into categories, tillage influenced broadleaf weeds in 1994 and 1996 and total weeds in 1995. Plots under the NT regime had an average of 46 broadleaf weeds m−2 compared to 27 in CT in 1994, with Amaranthus rudis Sauer (common waterhemp) being the most prevalent. NT had a total of 186 weeds m−2 compared to 125 m−2 weeds in CT in 1995; however, in 1996, CT plots had 184 weeds m−2 compared to 121 m−2 in the NT regime. Except for broadleaf weeds in 1994, crop rotation did not influence the number of weeds, and herbicide application methods had the greatest effect on weed populations. Overall, weed populations were greater in 1997, 1996, and 1995 than in 1994 for all herbicide application methods. The no-herbicide treatment had the highest number of weeds throughout the study. The total number of weeds in band and broadcast treatments averaged 41 and 26 m−2 in 1994; 96 and 24 m−2 in 1995; 96 and 12 m−2 in 1996; and 109 and 95 m−2 in 1997. The use of broadcast herbicides in NT should be recommended for land coming out of CRP. Regardless of the herbicide application method or crop rotation, CT plots had better yields for both Zea mays L. (corn) and Glycine max L. (soybean). Glycine max had a better stand compared to Z. mays in the first year, indicating that a rotation starting with G. max might be preferred in the land coming out of CRP.

The effect of storage conditions on longevity of crenate broomrape seed was determined and then used to predict and simulate new infestations. Newly harvested broomrape seed mixed with soil and placed in nylon mesh bags was buried in soil 8 and 23 cm deep or kept at room temperature in the laboratory for 6 yr. In the field, seed exhibited a seasonal pattern in germination, showing consistent and poor germination from September to January and for the rest of the year, respectively. The two burial depths tested showed no significant differences in seed germination or survival. Seed stored under laboratory conditions germinated throughout the year. The negative exponential model (% germination = A exp−-B-months of storage) described the time course of germination of crenate broomrape seed. Seed longevity decreased much quicker in the field than in the laboratory. For example, germination capacity decreased 66 and 95% and only 27 and 60% after 33 and 108 mo of soil burial or under laboratory conditions, respectively. According to these findings, the repeatedly reported infestations occurring in plots after many (up to 14) years of not growing broomrape-susceptible crops can not be attributed to the longevity of buried seed.

Glyphosate is the world's most widely used herbicide. It is nonselective and has been used to control a broad range of weed species for the past 20 yr, without the appearance of resistant weed biotypes. However, a biotype of Lolium rigidum from a field in Northern Victoria, Australia, in which glyphosate had been used for the past 15 yr, failed to be controlled by label recommended rates. Based on LD50 values from pot dose-response experiments, this biotype exhibited resistance to glyphosate and was nearly 10-fold more resistant compared to the susceptible biotypes tested. The biotype was resistant to three different salts of glyphosate. The biotype was also nearly threefold more resistant to diclofop-methyl but was susceptible to other commonly used selective and broad-spectrum herbicides. Between the two-leaf and tillering stages of development, a susceptible biotype exhibited a small but significant decrease in tolerance to glyphosate, whereas tolerance of the resistant biotype remained unchanged with age. The resistant phenotype was verified in experiments in which seed was germinated in the presence of glyphosate. Observations on shoot and root growth of seedlings in these experiments suggested that the resistance mechanism might be associated more with the shoot than with the root.

Kyllinga species are becoming more common throughout the southeastern United States. Two species, Kyllinga brevifolia and Kyllinga squamulata, in particular are prevalent weeds in turfgrass. To better understand these weeds, growth chamber studies determined the growth of K. brevifolia, K. squamulata, and Cynodon dactylon × Cynodon transvaalensis as influenced by three temperature regimes (33/24, 25/17, 19/11 C day/night, respectively). Temperature influenced almost all aspects of Kyllinga species growth. Plant height of both Kyllinga species increased nearly twofold after 8 wk at high temperatures. Plants were mowed each week to 2.5 cm; both species produced more than twice as many clippings by 8 wk at high (33/24 C) temperatures than at low (19/11 C) temperatures. Destructive analysis at 8 wk revealed that K. brevifolia shoot and root weight increased with decreasing temperature, whereas K. squamulata shoot and root weights were not affected by temperature. Shoot weight percentage for both Kyllinga species increased from 59% in medium temperatures to 69% in high temperatures. K. brevifolia shoot weight percentage decreased to 53% in low temperatures, whereas K. squamulata shoot weight percentage increased to 72%. K. brevifolia inflorescences formed at 2, 3, and 5 wk in high, medium, and low temperatures, respectively, whereas K. squamulata flowered immediately in all temperatures. C. dactylon × C. transvaalensis and Kyllinga species growth were similar within each temperature regime throughout the 8-wk study.
