Axes, hypocotyls and radicles excised from dry cotton seeds (Gossypium hirsutum L. cv. M-8, a double haploid) were imbibed for 24 h and compared with axial segments (excised sections of embryos below the cotyledons) of imbibed, intact seeds. Radicles of excised axes had a 7.4-fold increase in length compared with only 5.2- and 5.7-fold increases, respectively, in radicles of intact seeds and in those isolated when dry. Change in hypocotyl length was not as extensive. EM data for hypocotyl and radicle cortical cells from dry and imbibed seeds revealed a major reduction in matrix protein in protein storage vacuoles along with significant organelle development at 24 h from the start of imbibition. This occurred in parallel with a reduction in salt-extracted proteins and an increase in 2% SDS-extractable proteins. SDS-PAGE of protein from low (0.2 M NaCI) and high (1.0 M NaCI) salt extracts showed a reduction in amount of the major storage proteins (53 and 48 kDa), these bands being almost totally absent in gels of protein extracts from imbibed radicles and significantly reduced in hypocotyls, within 24 h from the start of imbibition. These results indicate that initial elongation of hypocotyls and radicles in intact seeds or of excised axes, after 24 h imbibition, involves breakdown of storage proteins in these axial parts to supply nutrients for growth, with very limited contribution from the cotyledons.

The seed coat of Trifolium repens L. was studied with special emphasis on the development of the strophiole, which is the site for water entry during imbibition in leguminous seeds. The epidermal cells of the strophiole are longer than the cells in the remainder of the seed epidermis in the mature ovule. During seed development the median cells of the strophiolar epidermis divide periclinally into an outer layer of palisade cells and an inner layer of isodiametric cells. Prior to maturity a fissure is formed between some of the palisade cells in the centre of the strophiole. It is suggested that tension develops between the palisade cells and the iso-diametrical cells during later maturation stages causing the formation of the fissure which it is believed functions in water uptake. It is indicated that the ‘light line’ is caused by alteration of cellulose microfibrillar orientation in palisade cell walls. It is confirmed that removal of the epicuticular wax from hard seeds by rinsing in absolute alcohol or hexane does not induce water imbibition. Only when seed coats are mechanically abraded do hard seeds germinate.

Experiments were performed to determine if RAPD profiles developed using total DNA isolated from soybean seed could be affected by the physiological state or the quality of the seed. RAPD profiles were developed using template DNA isolated from a single seed lot of soybean (Glycine max L. cv. Kirby). High quality seeds were used to produce four populations varying in either quality or physiological state: untreated control seed ambient temperature and humidity storage for 12 months, accelerated aging at 41°C and 100% relative humidity for 48 h, and controlled hydration (seed priming). One hundred and eighty-eight primers were used to create separate RAPD profiles from total DNA isolated from each set of seed and from soybean leaf tissue. Sixteen polymorphisms from 14 primers were identified as a result of seed treatments. Six primers showed nine polymorphisms in RAPD profiles of ambient-stored seed DNA, while four and two primers produced polymorphisms in reactions using accelerated aged or primed-seed template DNA, respectively. Two primers showed a polymorphic fragment in vegetative DNA not observed in any of the seed DNA samples. Ten of the observed polymorphisms were due to the appearance of a DNA fragment in response to a specific seed treatment while six were the result of the treatment-induced loss of a DNA fragment. The six polymorphisms resulting from the loss of a major fragment were all due to ambient-temperature seed storage. Results were reproducibly obtained from multiple DNA isolations using three separate DNA isolation procedures involving either multiple seed or a single seed as the template source. Therefore, genetically identical seed can consistently display RAPD polymorphisms as a response to the environmental exposure.

Benzyladenine, a synthetic cytokinin, stimulates DNA metabolism during maize germination; apparently such stimulation occurs in two phases, and during the first phase stimulations of DNA synthesis, of protein kinase activity (both α-amanitin repressible), of DNA polymerase and poly ADP-ribosyl polymerase activities are observed. Since the evidence suggested that the effects produced by BA were due, at least partially, to transcriptional activation, attempts were made to discover genes, by subtractive hybridization of a cDNA library or by differential display, whose expression depended on the presence of BA; however these were unsuccessful. BA modulated the expression of a limited number of genes only. The possible mechanism by which BA could be promoting DNA metabolism during maize germination is discussed.

An element of storage stability of many orthodox seeds is that the embryo cytoplasm is vitreous at normal storage temperatures. That is, as drying proceeds during seed maturation, intracellular solutions become so concentrated and viscous that diffusional movement is all but eliminated (Williams and Leopold, 1989). In this report the relationship between desiccation tolerance and the ability to form the vitreous state as germination proceeds is examined. As pea seeds (Pisum sativum cv. Alaska) imbibed water up to 18 h, no change in the glass transition temperature was seen, and the embryos remained desiccation-tolerant. Between 18 and 44 h of imbibition, the axes lost the ability to exhibit a distinguishable vitreous transition, and the embryos had lost the ability to survive desiccation. After about 50 h, embryos again showed vitrification but only at markedly lower temperatures, as would be expected to accompany the loss of oligosaccharides (sucrose, raffinose, stachyose and verbascose) and their replacement by monosaccharides during early germination (Koster and Leopold, 1988). Thus, the loss of desiccation tolerance during germination in pea seeds appears to be associated with a loss of the high temperature oligosaccharide: water glass and a subsequent appearance of a new glass transition at a lower temperature resulting from the accumulation of monosacchrides.

Dormant weed seeds were germinated at constant and alternating temperatures using five chemicals. Germination of Chenopodium album when expressed in normal equivalent deviates, increased linearly with the logarithm of chemical dose up to an optimal concentration. Ethephon (2-chloroethyl phosphonic acid) and sodium azide slightly stimulated germination of C. album seeds at 20°C while potassium nitrate, thiourea and azide each interacted positively with alternating temperatures (5°/25°C). Relief of Avena fatua dormancy by azide exhibited a positive interaction with cool constant temperatures (3–10°C), and chemicals had smaller or no effects at higher and alternating temperatures. No chemical mixture, including that of nitrate, thlourea, ethephon, azide and hydrogen peroxide, promoted more germination than azide on its own in A. fatua or nitrate together with ethephon in C. album. In C. album, this result arose largely because nitrate and azide antagonized each other in the presence of ethephon. Peroxide and thiourea together relieved this antagonism so that the fourth order interaction was positive. The negative interaction of nitrate, azide and ethephon in C. album was avoided in a sequential treatment with nitrate plus ethephon for seven days followed by sodium azide. Relief of dormancy by nitrate and ethephon was optimized at 67 mM nitrate with 1.4 mM ethephon for C. album at 20°C and 20 mM nitrate with 1.4 mM ethephon for A. fatua at 15°C. None of the chemicals used in this investigation stimulated the germination of Rumex crispus seeds. The results are discussed with respect to chemical control of soil seed banks.

Amounts of mineral nutrients and aluminium (Al) were assessed in the globoid inclusions, proteinaceous matrix and druse crystals of Eucalyptus calophylla seeds collected from trees grown in coal-mine (mean soil pH 4.3, Al 260 μg g−1) and forest soils (pH 5.3, Al 10 μg g−1). Energy-dispersive X-ray microanalysis (EDX) of bulk frozen hydrated samples indicated that significantly higher amounts of Mg, P, S, K and Ca occurred in the globoid inclusions of mine- than forest-site seeds. In both seedstocks, Al was detected in the globoid inclusions but not in the proteinaceous matrix or druse crystals. Significantly higher amounts of Al occurred in mine-site samples of dry and germinating seeds than in forest-site seeds. It was concluded that globoid inclusions may be useful as indicators of the soil conditions in which the parent plant was grown.

A partial amino acid sequence of a vigour-related protein, V-1, from cauliflower (Brassica oleracea L.) seeds was obtained, after isolation by two-dimensional gel electrophoresis and gas-phase micro-sequencing. The sequence was found to have high homology to soybean seed maturation proteins. However, the position of the sequence in the V-1 polypeptide suggests that the V-1 and soybean proteins have different polypeptide structures. An osmoprimed seeds cDNA library was constructed and has been screened by a cDNA probe derived from the V-1 sequence. Two cDNA clones with high homology to aspartic protease (EC 3.4.23) from barley grain were isolated. The expression of putative aspartic protease mRNA was found to be enhanced by osmopriming, however the clones were found to have no significant sequence homology to the V-1.

The endosperm and cotyledon tissues of Ricinus communis seeds and young seedlings were examined for changes in the mineral nutrient composition of globoids during early seedling growth. The effect on globoid composition of providing mineral nutrients to the developing seedling was also investigated. Globoids in endosperm and cotyledon tissues of castor bean seeds contained P, Mg and K, as well as trace amounts of Ca, Fe and Zn. Irrespective of the addition of mineral nutrients, K content in globoids of endosperm and cotyledon tissues declined significantly during initial seedling growth. During early seedling growth, amounts of Fe, Zn and Ca increased in cotyledon globoids. Ca contents of globoids of endosperm tissues also increased. The changes in Fe, Zn and Ca globoid contents were not influenced by providing mineral nutrients to growing castor bean seedlings.
