An ultrastructural examination of Fellodistomum fellis has revealed that the caecal lining consists of 2 distinct components: (a) cup-shaped digestive cells and (b) a pleomorphic layer of cytoplasm which supports and separates individual digestive cells. The digestive cells sequester host blood components within apical pockets formed by lamellated extensions of the cell surface, and undergo asynchronous, cyclical transformations in morphology associated with extracellular digestion and with the extrusion to the gut lumen of pigmented digestive residues. Histochemical tests and elemental analysis of the pigment suggest that it is a ferripor-phyrin, haematin. In the anterior portion of the caecum the supporting cytoplasmic layer is in continuity with the oesophageal tegument and snares the same ultrastructure. It is concluded that the digestive cells are supported by an extension of the foregut tegument.

Guppies (Lebistes reticulatis) exposed to doses as low as 25 cercariae of Diplostomum spathaceum for 30 min were consistently and significantly more susceptible to predation by brook trout (Salvelinus fontinalis) than uninfected controls. The increase in susceptibility to predation was correlated with a marked decrease in the activity of infected fish.

At concentrations ranging from 0·1 to 10·0 mg/l, copper and zinc reduced both the longevity and infectivity of Echinoparyphium recurvatum cercariae. Concentrations of 10·0 mg/l copper and zinc in hard water reduced the time to 50% mortality of cercariae from 30·5 h to 8·5 h and 15·5 h respectively. Copper-induced effects upon cercarial infectivity were particularly severe and exposures of cercariae to 0·5 mg/l of this metal for as little as 15 min caused significant reductions in their ability to infect molluscan 2nd intermediate hosts. Water hardness had a marked influence on copper toxicity but had a much lower effect on the toxicity of zinc. Metal concentrations found to exert a profound influence on parasite transmission in the present laboratory-based study have been found to occur in natural, albeit polluted, freshwater habitats.

Colour changes that occur during tanning of intact and punctured Globodera rostochiensis females and of isolated cuticle are described. Experiments with catechol and enzyme inhibitors show that the tanning enzyme, a phenoloxidase, is located in the cuticle, soluble in aqueous media and relatively abundant. Substrate for the enzyme is limited in the cuticle. Pseudocoelomic fluid of yellow females promoted tanning and contains either phenolic substrate(s) for the enzyme or part of a biochemical system responsible for release of the substrate. The substrate or biochemical system is thermolabile. Apparently, the tanning enzyme and its substrate(s) are partitioned in living females but the partitioning breaks down after death and the cuticle tans to form the cyst.

The distribution of exposed and hidden lectin-binding glycoconjugates in adult Schistosoma mansoni worms was studied. Fluorochrome-conjugated lectins were allowed to react both with intact worms and with frozen sections of worms. The results show a selective reactivity of lectins with tegumental surface structures, the intestine, eggs and basement membranes. Strong binding of wheat germ agglutinin (WGA) was seen to the exposed surface of intact worms. Peanut agglutinin (PNA), Ricinus communis agglutinin (RCA) and concanavalin A (Con A) gave a weaker reaction. There was a preferential affinity of PNA and RCA to the spikes covering the tubercles of male worms whereas Con A gave a more uniform staining pattern. Internal structures exposed in frozen sections of worms gave distinct staining reactions. Binding sites for Con A were widely distributed throughout the worm. PNA produced a more selective binding and was the only lectin staining basement membranes as distinct structures. There was a strong selective staining of the gut with soybean agglutinin (SBA) and RCA. Eggs seen in large quantities inside female parasites reacted strongly with SBA and PNA and less intensely with RCA and WGA. The observed selectivity of lectin binding may reflect physiological function. The results may be important as a basis for using lectin affinity-chromatography for purification and characterization of worm constituents

The level of immunity to a percutaneous cercarial challenge with Schistosoma mansoni was assayed 4–6 weeks after immunization of mice with highly irradiated (20 krad.) cercariae or schistosomula. When immunization and challenge occurred through the same skin site, resistance, particularly that which occurred in the skin, was greater than that observed when immunization and challenge occurred in different sites. The enhanced resistance is believed to be due to localized changes in the skin; 4 weeks after exposure to irradiated cercariae, abdominal skin is characterized by a thickened epidermis, changes in the ground substance and a cellular infiltration of the dermis. A convenient mouse model is described in which one or both ear pinnae are exposed to irradiated cercariae and a percutaneous challenge is given via the abdomen, thus eliminating the effects of local skin changes. In this model, the majority of the challenge infection which succumbs to the immune response appears to be killed in the skin.

F1, Quackenbush (Q) × Wild (W) Mus musculus were assayed and selected for their level of immunity to secondary infections with 100 Nematospiroides dubius using faecal parasite egg counts. A significant positive linear correlation was found between the number of parasite eggs voided in the faeces and the number of N. dubius recovered after secondary infections in mice. Selection for immunity was not influenced by the conformation of the mice. High (H) and low (L) responder colonies together with a randomly (Rd) bred colony of mice were established over 5 generations. Adaptive immunity was more efficacious in protecting female than male mice and appeared to be controlled by a few dominant genes; the heritability (h2) of the trait was estimated at 0·56 for the parental stock and 0·55 for the F5 Rd colony and the realized heritability was 0·49. Higher anti-N. dubius antibody titres were detected in H than L mice. H mice harboured fewer and less fecund parasites than Rd colony mice; L mice had more worms with greater fecundities than Rd mice. The levels of primary infection in H and L mice related to the levels developed after secondary infection from F4. This suggested at least partial linkage of genes controlling innate with genes controlling adaptive immunity against N. dubius.

Trypanosoma brucei variable surface glycoproteins (VSGs), isolated from some antigenic types of trypanosomes, degraded during isolation. We show that this degradation occurred immediately after breakage of the organism, presumably because of liberation of internal enzymes, and resulted in heterogeneity of isolated VSGs with respect to charge and/or molecular weight. Degradation and consequent heterogeneity of these VSGs could be abolished by releasing VSG from the trypanosome surface without breakage of the organism. The method was a modification of an incubation and shaking procedure initially described for Trypanosoma congolense (Reinwald, Rautenberg & Risse, 1979). WaTat 1.11 VSG released by this method and isolated, had a molecular weight (mol. wt) of 63000 and bound to a heterologous anti-VSG serum. VSG isolated from the same trypanosomes following breakage of organisms was of lower molecular weight. One such WaTat 1.11 VSG fragment of 42000 mol. wt, did not bind to the heterologous anti-VSG serum and therefore lacked cross-reacting antigenic determinants present in the 63000 mol. wt VSG

The intestinal flagellates, Giardia muris and Spironucleus muris, cause similar infections in CBA mice as determined by trophozoite and cyst counts. Both parasites occur all along the small intestine with G. muris, being mainly present in the anterior part and S. muris towards the posterior. The early stages of infection are similar in all strains of mice examined and peak levels of both trophozoites and cysts occur 1–2 weeks after infection. All strains of mice overcome the infection but the rate of recovery varies considerably between strains, being most rapid in BALB/c and slowest in A and C57BL.B10. Outbred mice are more variable in their recovery than inbred mice. After recovery, mice are partially resistant to reinfection with the homologous but not the heterologous parasite. Resistance to reinfection with S. muris is greatest in those strains that eliminate the primary infection most rapidly. Giardia muris and S. muris cause similar changes in the mucosa of the small intestine of BALB/c mice with increased intra-epithelial lymphocyte counts from 3 weeks onwards corresponding with the start of the elimination of the parasites from the gut. A reduction in villus height and increase in crypt depth is also characteristic of these infections.

The influence of the late components of complement (C) on percutaneous primary infections with Schistosoma mansoni was studied in inbred mice genetically deficient or intact in C5. Worm recoveries were diminished in C5-deficient male mice as compared to C5-intact animals. Twenty-four-day-old parasites were also shorter following growth in C5-deficient animals. At 7 weeks after infection, female schistosomes, but not male parasites, were shorter and fewer eggs/schistosome pair were deposited in the livers of C5-deficient as compared to C5-intact male mice. In addition to the presence of C5, the mouse sex was found to influence the outcome of an infection. Schistosomes showed a reduced infection rate and were relatively stunted after 24 days of growth in female mice of the two strains as compared to male mice of the corresponding strains. The results suggest that C5 plays no role in defence against a primary infection in mice.

The surface topography of the tegument of adult Schistosoma mekongi was studied by scanning electron microscopy (SEM). In comparison to other species of human schistosomes the male tegument lacks tubercles and, except in the gynecophoral canal, also lacks spines; instead the surface is composed chiefly of trabeculae of highly perforated ridges which give it the ‘spongy’ appearance. In addition, there are 3 kinds of papillae interspersed on the surface among the ridges. The first is a doughnut-shaped papilla with a central crater which is most abundant on the ventral surface of the anterior part, on the floor of the gynecophoral canal and on the dorsal-lateral aspect of the tail. The second is a pleomorphic papilla with irregular shape and size, which is scattered throughout the dorso-lateral aspect of the middle part of the body. The third type of papilla has a uniform hemispherical shape, possesses a cilium projecting from its apex and probably corresponds to the ‘sensory papilla’ found in other species. The tegument of the female differs from that of the male by having numerous short spines over the whole surface; however, the pleomorphic papillae are much fewer in number and the ridges are much less developed than those of the male tegument; complex trabeculae are absent.

In mice infected with the intestinal flagellates Giardia muris or Spironucleus muris, together with the blood parasites Babesia microti or Plasmodium yoelii, there is a temporary decrease of flagellate cyst output coincident with the peak of the blood parasite infections, followed by a rapid return to normal levels. This decrease in cyst output is correlated with decreased numbers of trophozoites in the small intestine. The effect on S. muris is more marked than that on G. muris. Neither blood parasites has any effect on the total duration of the flagellate infection and the flagellates do not affect the blood parasites. In mice infected with G. muris or S. muris and P. berghei there is also a decrease in cyst output but this is less apparent than in infections with B. microti or P. yoelii because of the fatal nature of the P. berghei infection. It is suggested that the decrease in cyst output is probably due to changes in the contents of the small intestine or to non-specific immunological factors rather than to specific immunological changes.

Six murine monoclonal antibodies to Leishmania tropica major have been prepared and the properties of these antibodies studied. Two (WIC 79.3 and 79.7) were L. tropica major species-specific and bound to promastigote cell surfaces, to parasitized macrophages, but not isolated amastigotes. No evidence was found for the production of antibody to the antigenic determinants recognized by WIC 79.3 or 79.7 during L. tropica major infections in mice and hamsters. One antibody (WIC 79.1) bound to sub-cellular organelles of Leishmania species but to a different sub-cellular organelle of Trypanosoma cruzi. Two others bound to the flagellum, one (WIC 79.2) to all Leishmania species, T. cruzi and Trypanosoma brucei, the other (WIC 79.4) only of L. tropica major and L. donovani species. One antibody (WIC 79.5) was directed against an unknown internal antigen found in all Leishmania species and T. cruzi

Unsuckled mother rats given a 1 h suckling stimulus 3 h after subcutaneous injection of an exact dose of homogonic Strongyloides ratti allow fewer worms to develop in their intestines by day 8 than nulliparous rats (Wilson & Simpson, 1981). This effect is studied in more detail in terms of the length of time between weaning and stimulus (W → S) and injection and stimulus (I → S). It was observable with a W → S of 30 h but this and a period of 5 h were less effective than 24 h. With W → S constant at 24 h, significantly more worms developed in mothers when I → S was 24 h compared to 3 h and 10 h (P < 0·005). The data, combined with those from nulliparous controls, are presented as a measure of the change with time of numbers of larvae in that compartment of the system which gives access to the stimulated mammary gland. It is argued that the particular compartment is the local lymph node draining the injection site and that the kinetics deduced are applicable to migration in the rat in general

The antigenic differences and similarities between culture forms of Trypanosoma (Schizotrypanum) hedricki, Trypanosoma (Schizotrypanum) myoti, Trypanosoma (Schizotrypanum) vespertilionis and Trypanosoma (Schizotrypanum) dionisii, all isolated from bats, were compared using the double-diffusion technique. Antiserum to each trypanosome species was produced in rabbits by inoculating them with sonicated culture forms. Two isolates of T. hedricki were antigenically identical to each other, as were 2 isolates of T. myoti. There were several common antigens between the species. However, at least 1 antigen of each species was distinct from those of the other species.

Development of surface topography of cultured Schistosoma mansoni was followed in the cercariae, newly transformed and older schistosomula, and adults. Comparison with surface features of schistosomes from animal infections revealed no apparent differences in tegumental folding or morphogenesis, spines, sensory structures, or other components. Eggs produced by worm pairs grown in vitro were grossly abnormal, usually composed of loosely packed vitelline granules and lacking a shell.

We report the results of experimental work on (a) the influence of temperature on the age-dependent survival and infectivity of the mira-cidia of Schistosoma mansoni and (b) the relationship between snail age, snail size and susceptibility to infection. The death rate of miracidia declined exponentially with age where life-expectancy was maximal (approximately 16 h) at 15 °C. Infectivity also declined rapidly with larval age but, in contrast to larval survival, the rate of infection was at a maximum at 25 °C. Snail susceptibility was shown to be more closely correlated with host size rather than host age. Susceptibility declined exponentially with increased host size. Size-dependent susceptibility was shown to generate concave age-prevalence curves for infection within snail populations, where the maximum prevalence is generated in snails of intermediary age. Simple mathematical models are developed to aid estimation of larval survival and infection rates and experimental results are discussed in relation to the overall transmission success of the parasite from man to snail.

A total of 120 young cotton rats were fed ad libitum for 10 weeks on either synthetic diets containing 2·5, 5, 10 or 15% casein or on a stock breeding diet containing 20% crude protein. Half of the animals in each group were infected with the filarial nematode Litomosoides carinii and the food intake and body weight of each animal was measured. There was a trend towards increased food consumption in protein-deficient cotton rats and this was increased further in infected animals; however, differences between groups were not significant. Low protein diets reduced the rate of growth of uninfected and infected cotton rats; filarial infection intensified these adverse effects of under-nutrition and increased the number of rats which died in the lowest protein group. Protein deficiency had little effect on the number of L. carinii which developed, although fewer parasites became established in cotton rats fed on a 2·5% diet than in those fed on a 10% diet. Parasites developing in protein-deficient animals were shorter than those developing in well-fed ones and embryo-genesis was retarded in female worms from protein-deficient animals. The onset of patency was retarded in protein-deficient animals and the microfilaraemia which developed subsequently was lower up to 120 days post-infection.

Since the in vitro production of trypomastigote stages of Trypanosoma cruzi in cell culture is frequently limited by (1) host cell overgrowth and (2) by the unequal redistribution of parasites after cell division resulting in asynchronous release of trypomastigotes, a culture system was devised in which host cell mitosis was inhibited by irradiation prior to parasite infection. L-6 rat myoblast cells when exposed to 3000 rad. of gamma radiation lost their ability to divide but remained susceptible to infection with, and capable of supporting the intracellular growth of, T. cruzi. Using this approach it proved possible to have virtually 100% of cells infected and achieve much better synchronization of trypomastigote release than with conventional culture systems. Additionally, the total number of parasites provided approached 1 × 109 trypomastigotes/ 150 cm2 flask, a significant increase over other culture systems. Preliminary studies with Plasmodium fallax and Eimeria tenella indicate that irradiated host cells may be utilized to advantage for the cultivation of other intracellular protozoa.

The relationship between infection success, the density of Schistosoma mansoni miracidia and the density of the snail host Biomphalaria glabrata was examined experimentally. Within laboratory infection arenas, the distribution of miracidial infections/snail was approximately uniform and the net rate of snail infection was directly positive binomial to miracidial density. In contrast, however, the rate of infection declined exponentially as snail density increased. Mathematical models are developed to aid in the estimation of infection rates and experimental results are discussed in the context of the transmission dynamics of schistosomiasis.
