Milk fat was fractionated into liquid (m.p. ⋍ 12 °C), intermediate (m.p. ⋍ 21 °C) and solid (m.p. ⋍ 39 °C) fractions by three different processes—melt crystallization, short-path distillation and supercritical CO2 extraction—and the cholesterol content of these fractions determined. Cholesterol was enriched in the liquid fractions from all three processes, in particular about 80% of the cholesterol being found in the liquid fraction obtained by short-path distillation. The basis of migration of cholesterol into various milk fat fractions was explained by its affinity to various triglycerides (melt crystallization) and by vapour pressure and molecular weight (short-path distillation). It was more complex in the supercritical CO2 extraction process; the interplay of cholesterol affinity toward CO2 and its molar volume, and its vapour pressure enhancement under applied pressure play a role.

Changes in microbial flora during the cheese-making and maturation processes of Spanish mould-ripened Cabrales cheese have been studied. In the interior of the cheese lactic streptococci, mainly Streptococcus lactis, predominated before and during salting (0–4 d); Str. lactis and lactic acid-utilizing yeasts during dryingroom ripening (5–15 d); moulds, mainly Penicillium roqueforti, and lactobacilli, mainly Lactobacillus plantarum, predominated during cave-ripening (16–120 d). On the cheese surface Str. lactis was the majority flora before and during salting; Str. lactis, micrococci and leuconostocs were the main microbial groups during drying-room ripening, and micrococci throughout cave-ripening. Coliforms and coagulase-positive staphylococci died off during the first month of maturation.

The growth in wirelessly enabled sensor network technologies has enabled the low cost deployment of sensor platforms with applications in a range of sectors and communities. In the agricultural domain such sensors have been the foundation for the creation of decision support tools that enhance farm operational efficiency. This Research Reflection illustrates how these advances are assisting dairy farmers to optimise performance and illustrates where emerging sensor technology can offer additional benefits. One of the early applications for sensor technology at an individual animal level was the accurate identification of cattle entering into heat (oestrus) to increase the rate of successful pregnancies and thus optimise milk yield per animal. This was achieved through the use of activity monitoring collars and leg tags. Additional information relating to the behaviour of the cattle, namely the time spent eating and ruminating, was subsequently derived from collars giving further insights of economic value into the wellbeing of the animal, thus an enhanced range of welfare related services have been provisioned. The integration of the information from neck-mounted collars with the compositional analysis data of milk measured at a robotic milking station facilitates the early diagnosis of specific illnesses such as mastitis. The combination of different data streams also serves to eliminate the generation of false alarms, improving the decision making capability. The principle of integrating more data streams from deployed on-farm systems, for example, with feed composition data measured at the point of delivery using instrumented feeding wagons, supports the optimisation of feeding strategies and identification of the most productive animals. Optimised feeding strategies reduce operational costs and minimise waste whilst ensuring high welfare standards. These IoT-inspired solutions, made possible through Internet-enabled cloud data exchange, have the potential to make a major impact within farming practices. This paper gives illustrative examples and considers where new sensor technology from the automotive industry may also have a role.

The rennet coagulation times of infant milk formulae or fresh skim milk (milk) samples heated at temperatures in the range 70–140 °C for 1–10 min decreased on acidification, usually to pH < 6·0. Heated milk samples acidified to pH 5·5 and reneutralized to pH 6·6 retained good rennet coagulability. Acidification of such milk samples before heating also reduced the adverse effect of severe heat treatment (95 °C for 1 min) on rennet coagulation. Addition of low concentrations of CaCl2 to heated milks offset the adverse effects of heating. Acidification of heated milks increased the [Ca2+], and reneutralization of acidified milk only partly restored the [Ca2+], i.e. acidified/reneutralized milk had a higher [Ca2+] than normal milk, suggesting this as the mechanism via which acidification/neutralization improves the rennet coagulability of heated milk. Approximately 50% of the whey protein can be incorporated into rennet gels in heated milks while retaining good coagulability and curd tension; this may be a useful technique for increasing cheese yield.

1. A study of the compositional factors which affect the heat coagulation of milk has been undertaken.

2. Calcium-ion concentration and colloidal phosphate content appear to be the chief factors determining the tendency of a milk to coagulate on heating.

3. Acidity (mainly derived from thermal decomposition of lactose and casein) and heat denaturation of casein are supplementary coagulation factors which develop during the heating process.

4. Lactose, as the main source of heat-developed acidity, is an important secondary factor in heat coagulation, but not an essential one. Coagulation can proceed, though more slowly, in its absence. The serum proteins play no part in the phenomenon.

5. A provisional theory of the heat coagulation of milk based on these findings is put forward.

Melting curves were determined by differential scanning calorimetry on 17 samples of milk fat obtained at approximately fortnightly intervals through one dairying season. Liquid-fat content at a number of temperatures was calculated from the melting curves.

Correlation between percentage liquid fat at 12 and at 22 °C and weight percentage of short-chain fatty acids (C4–C8) was low (r = 0·45 and 0·58 respectively) as was that with cis-unsaturated acids (r = 0·39 and 0·29). Correlation with the sum of short-chain and cis-unsaturated acids was higher (r = 0·71 and 0·68). With a multiple regression model including short-chain and cis-unsaturated acids the values of the multiple correlation coefficient (R) were 0·75 and 0·78.

The softening point showed a high correlation (r = 0·79) with weight percentage of long-chain saturated acids and also with the temperature at which 94 % of the fat was liquid (r = 0·75).

Possible explanations of these findings are discussed.

A quantitative method for rapid routine analysis of individual free fatty acids (FFA) in milk was developed. Lipid was extracted from milk in ether and FFA were recovered by shaking the extract with anion exchange resin Amberlyst 26. The resin-bound FFA were methylated directly and the individual acids quantified, using internal standards, by gas-liquid chromatography. The properties of the resin were measured. The validity of the method was established by extraction of FFA mixtures and milk. Individual acids were, on average, found to be within 6% of the actual concentration present in the mixture. An average coefficient of variation of 4·3% was achieved for the major individual fatty acids on repeated extraction of a single milk sample.

A sample of milk fat was fractionated into high, medium and low molecular weight triglycerides by silicic acid column chromatography. Each fraction was further separated by silver ion adsorption thin-layer chromatography into triglyceride classes which were subjected to stereospecific analysis. Trans-monoene fatty acids were incorporated into triglycerides in the same manner as their cis-isomer. The specific positional distribution for fatty acids in intact milk fat triglycerides was not always maintained in the molecular weight fractions or in the various classes of unsaturation. The significance of these findings is discussed in relation to biosynthetic mechanisms for milk fat synthesis.

Two methods were compared for the determination of free fatty acids (FFA) from acetic to long-chain acids in samples with a large excess of triacylglycerols (TG) (1[ratio ]200, w/w), such as cheese and other dairy products. In method 1, after fat extraction, FFA were separated from TG by aminopropyl-bonded phase chromatography, injecting the fraction containing FFA directly into the gas chromatograph. In method 2, extracted fat was treated with tetramethylammonium hydroxide, the methyl ester derivatives being formed in the injector. Cheese samples and standard mixtures of FFA and TG in different proportions were analysed by both methods. The cheese sample contained 2·4 times more FFA when analysed by method 2 as compared with the result obtained with method 1. The composition of the standard mixtures analysed by method 1 closely reflected that of the original mixture and gave 90–100% recovery of FFA, regardless of their chain length and the ratio of FFA[ratio ]TG (1[ratio ]1 or 1[ratio ]200, w/w). The composition of samples with a FFA[ratio ]TG ratio of 1[ratio ]200 (w/v) was severely distorted (as compared with the original composition of the sample) when analysed by method 2. Varying recoveries of FFA were also obtained, the largest differences being found for the shorter-chain components. We conclude that the FFA fraction should be separated from the TG fraction before derivatization and chromatographic analysis, particularly for samples in which the FFA represent a minor fraction of the TG.

Some strains of commercial cheese starters were obtained in higher yield after batch culture in whey-based media supplemented with soy peptones than in those supplemented with papain digested skim-milk. The soy peptones contained a greater proportion of small peptides than did the skim-milk digest. Also the average size of peptides was lower (3·4–3·9 residues) in soy preparations than in the milk digest (6·8). Small peptides isolated from the soy peptones were utilized by growing starters more completely than the equivalent peptides isolated from the papain digested milk. These observations may account for the difference in cell yields with soy and milk-digest peptones.

Twelve multiparous British Friesian cows in early (40±23 d in milk; n=6) or late (216±17 d in milk; n=6) lactation were used to study the effects of milking frequency on yield, udder volume and milk storage within the udder. After a 2 week control period of twice daily milking, diagonally opposed udder halves within a cow were milked once or twice daily for 3 weeks. Milk yield was 28–38% lower from the halves that were milked once daily than from halves that were milked twice daily. The loss of milk yield, expressed as a decrease in the relative milk yield quotient (an index that accounts for pretreatment differences), was greater for cows in early than in late lactation (0·59 v. 0·68). Empty udder-half volume was not decreased by once daily milking, suggesting that no cell loss occurred. Instead, once daily milking reduced the secretion efficiency (units of milk per unit of empty udder-half volume) by 46 and 27% respectively in early and late lactation; thus, at least part of the loss was due to reduced metabolic activity of the mammary epithelium. There were positive correlations between the relative milk yield quotient and the proportion (r=0·804) or volume (r=0·644) of cisternal milk in the glands that were milked once daily. These results confirm that, during extended milking intervals, milk loss was smallest for cows that stored a larger proportion of milk in the gland cistern.

The effects on the fatty acid composition of milk fat of feeding various oil-enriched diets (soya oil, red palm oil/palmitic acid and tallow) to dairy cows receiving a fat-deficient basal diet are reported. It was found that a linear relation was obtained between the dietary intake of C18 fatty acids and their output in milk fat. A similar relation, applicable only to the oil-rich diets, was found for the C16 acids. The significance of these relations is discussed. Fatty acid compositions and yields are discussed and compared with literature examples in which similar oil-rich diets were fed, but in which the low amount of fat in the basal diet did not markedly depress milk yield. The proportion of trans-isomer in the octadecenoic acid was found to increase appreciably on feeding soya oil, and to a lesser extent with dietary tallow. A linear relation between the 18:0 and 18:1 contents of milk fat was found, and its significance is discussed.

Micelles reconstituted from urea-treated milk by exhaustive dialysis against bulk milk were similar to native micelles with respect to colloidal phosphate: casein ratio, ethanol stability, heat stability and susceptibility to first-stage rennin action. Reconstituted micelles were considerably smaller than native micelles as indicated by turbidity, sedimentation and viscosity, had shorter second-stage rennet coagulation times, were unstable to [Ca2+] > 20 mM and had reduced base-binding capacity. It is suggested that the induced Ca sensitivity is due to unfavourable alterations in the micellar κ-casein coat arising from the decrease in average micelle size and can be offset by increasing the κ-casein complement of the system or by increasing the degree of aggregation by slowly raising the [Ca2+] level.

The antioxidant activity of casein and Maillard reaction products obtained by reaction of casein with glucose or lactose was studied. Antioxidant activity was evaluated in a model System containing methyl linoleate with haemoglobin as a pro-oxidant. Casein was antioxidative and heating casein in the presence of glucose or lactose resulted in enhancement of antioxidant activity. The development of antioxidant activity in reacted casein–sugar mixtures was determined as a function of initial casein and sugar concentration. The observed antioxidant activity of reacted casein–sugar mixtures was due to casein itself and Maillard reaction products resulting from reacting casein with sugar.

Plant oils in the diet are known to alter milk fat composition owing to changes in the supply of fatty acid precursors and/or activity of lipogenic enzymes in the mammary gland. Thirteen mid-lactating Alpine goats were used in a 3×3 Latin square design with 28-d periods to evaluate possible mechanisms regulating milk fat synthesis and fatty acid composition on grass hay-based diets containing none (H) or 55 g/kg diet dry matter of sunflower-seed oil (HSO) or linseed oil (HLO). Inclusion of oils in the diet had no effect on milk yield but enhanced (P<0·05) milk fat secretion. Compared with the control, HLO and HSO decreased (P<0·05) C10–C16 secretion and increased (P<0·05) C18 output in milk, responses that were accompanied by reductions in milk fat cis-9 14:1/14:0, cis-9 18:1/18:0 and cis-9, trans-11 18:2/cis-9 18:1 concentration ratios. Plant oil supplements decreased (P<0·05) mammary stearoyl-CoA desaturase (SCD) activity but had no effect on SCD mRNA. Treatments had no effect on glucose-6-phosphate dehydrogenase, malic enzyme and glycerol-3-phosphate dehydrogenase activity, or mRNA abundance and/or activity of lipoprotein lipase, acetyl-CoA carboxylase and fatty acid synthase in mammary, hepatic or adipose tissue. The results provided little support for milk fatty acid secretion responses to HLO and HSO being mediated via changes in mammary, hepatic or adipose mRNA abundance or in the activity of key lipogenic enzymes. In conclusion, plant oils in the diet enhance milk fat synthesis, alter milk fatty acid composition and specifically inhibit mammary SCD activity in the goat. Furthermore, the results suggest that the regulation of mammary lipogenesis in response to plant oils appears related to factors other than altered mammary gene expression or potential lipogenic enzyme activity.

Although there has been little study of the origin of intramammary infection (IMI) in goats, a common view is that most bacterial infection in goats occurs during milking. In the present study, the dynamics of occurrence of udder infection during and between lactations in three Anglo-Nubian goat farms in Israel was monitored. Coagulase-negative staphylococci were the predominant bacteria in the IMIs. We found that about 15% of the yearling does were already infected with bacteria when they joined the flock, whereas about 8% of the goats that dried-off returned with new IMIs. Moreover, virtually none of the goats acquired infection during lactation. Thus, our study showed that the aetiology of IMI in goats is very similar to that in dairy cows. A preventive treatment during the dry period should, therefore, be considered as an effective means of reducing the current rate of bacterial infections in goats.

Indigenous L-lactate dehydrogenase (LDH) in milk originates mainly from somatic cells, leucocytes and invading microorganisms. Its activity may be used for detection of mastitis. However, existing methods to measure LDH activity in milk both need pretreatment of the samples and still suffer from methodological problems. The present paper describes a fast, reliable method for determination of LDH activity, suitable for milk samples. The method is based on fluorometric determination of enzyme kinetics when L-lactate is converted to pyruvate. The assay uses raw milk without pretreatment and the method is easily adjustable to large-scale analyses on micro assay plates. Detection is based on (straight line) linear response within 4–7 min of initiation of the reaction. A substrate concentration of 35 mM in the reaction mixture was considered to be optimal for the assay. Intra plate assay precision was approx. 6% (CV) and the inter plate precision approx. 10%. Known inhibitors of LDH activity (oxidative direction), i.e., oxalic acid, oxamate, and pyruvate, were tested in different concentrations in order to verify the specificity of the response. The detailed kinetics of samples analysed indicated that the isoenzyme composition may have differed between milk samples, and that this composition may have been altered in high activity samples.

The heat stability of the milk supply to manufacturing creameries in south-west Scotland was examined over 15 months from Nov. 1975 to Jan. 1977. For all but 2 months of this period the heat stability of the milk was very highly significantly correlated with the naturally occurring level of urea. Between 72 and 90% of the variation of coagulation time (CT), measured at the natural milk pH, was accounted for by changes in milk urea alone. For a short period, in May and June, the CT of the milk at natural pH fell within the minimum of the CT–pH profile, but insufficient data were available to allow the occurrence of this phenomenon to be related to changes in milk composition.

A method has been developed for quantitative determination of para-κ-casein, involving spectrophotometric scanning of stained protein bands following polyacrylamide gel electrophoresis. The rate of hydrolysis of κ-casein in skim-milk at pH 6·6 and 30 °C was compared with that in EDTA-treated skim-milk under the same conditions. This showed that at the visually observed clotting time, at least 90% of the total κ-casein in milk had been hydrolysed. The time course of the reaction was consistent with all the κ-casein molecules being hydrolysed with the same efficiency. The results strongly suggest that essentially all of the κ-casein in milk is equally accessible to rennet action. This is consistent with the casein micelle being porous, or having all the κ-casein on the surface.

A method is described for determining H2S, methanethiol and dimethyl sulphide in Cheddar-cheese headspace. Cheeses ranging in age from 2 to 12 months were analysed using a gas chromatograph equipped with a photometric detector. These cheeses were presented to a trained flavour panel for the assessment of quality and intensity of Cheddar cheese flavour.

An attempt is made to correlate the concentration of sulphur compounds in the headspace with the average flavour scores. The results suggest that the flavour intensity of the cheese is related to the concentration of methanethiol in the headspace.
