The ripening-dependent changes in antioxidant activities and phytochemical content of mango (Mangifera indica L.) cultivar Safaid Chonsa at various ripening stages were evaluated. The ripening time period was divided into five stages (RSI-RSV) and the pulp was subjected to proximate analysis, antioxidant potential, and UHPLC/MS-based non-targeted metabolite fingerprinting. Proximate analyses depicted variations in moisture, dry matter, fat, protein, carbohydrate, and energy parameters. Maximum DPPH activity (51%) was observed at stages III, IV, and V while FRSP increased 31% at RS V as compared to stage I. Total antioxidant capacity and total reducing power potential were maximum (295.7 and 345.71 µg AAE/mg extract, respectively at stage V. Total phenolic content increased from 3.57 µg GAE/mg extract to 5.72 µg GAE/mg extract from stage I to RSIII while 19% increase in total flavonoid content was observed at stage V as compared to stage I. UHPLC/MS analysis showed presence of Aconitic acid, methylisocitric acid, 4-O-methyl gallate, beta-glucogallin, xanthenes, sakebiose, Isobergaptene, Fructoselysine 6-phosphate, Citbismine C, and many others at different ripening stages of chonsa mango extracts. The results conclude that during the mango ripening stages, changes in phytochemical composition have positive correlation with antioxidantive potential. These phytochemicals have nutritional and nutraceutical effects on human health therefore ripening stage should be considered for consumption of mango.

Echinococcosis lacks sensitive serological diagnostic tools. The echinococcosis-specific antigens Eg95, AgB8/1 and the Em18 gene sequences were fused and expressed as the novel recombinant antigens rAgB8/1-Em18-Eg95 (T3) and rEm18-Eg95 (T2), used for the diagnosis of hydatid disease, prepared into an enzyme-linked immunosorbent reaction (ELISA) kit, and evaluated for their serological diagnostic value. The relative molecular weight of the T3 protein was 88.1 kDa, the purified concentration was 1.5 mg mL−1, and the purity was 80%. The relative molecular weight of T2 protein was 79.9 kDa, the total protein concentration was 0.5 mg mL−1, and the purity was less than 50%. The overall coincidence rate of T2 protein was low, and it was impossible to distinguish between negative and positive sera. The T3 antigen was coated at 1.0 μg mL−1, the cutoff value was 0.5271, and the serum dilution ratio was 1:400. A T3 ELISA kits (96 tests) was constructed to detect the serum of 272 clinically and pathologically confirmed cases. The sensitivity of T3 was 93.8%, and the specificity was 83.3%. The parasite cross-reaction was 30%. Satisfactorily, the Pearson correlation coefficient between the T3 OD value and lesion diameter was 0.707, showing a strong correlation. T3 exhibits better antigenicity than T2, and the prepared T3 ELISA diagnostic kits reached the laboratory diagnostic level of a commercial kits. T3 can distinguish human cystic echinococcosis (CE) and alveolar echinococcosis (AE) more significantly and predict the diameter of lesions according to the OD value, which provides practical value for drug or surgical efficacy.

Clay fractions of eight vertisols and vertisolic soils from Israel were found to consist principally of a Fe-rich beidellite. Sediment volumes of Na-clay suspensions, obtained in measuring cylinders and read every 24 hr for as long as 720 hr, ranged from 3.8 to 8.4 cm/l00 mg clay and were as much as 19 times larger than corresponding suspensions of Ca-clays. Optical density data for all clay suspensions showed absorption curves typical of smectite. The relative number of platelets per tactoid, calculated from optical density measurements, ranged between 1.4 and 5.4 for the Na-clays and between 7.4 and 14.1 for the Ca-clays. In the Ca-clays, the sediment volume decreased with an increase in the relative number of platelets per tactoid. With increase in the major dimension of particles (calculated also from optical density curves), sediment volume tended to increase for the Na-clays and decrease for the Ca-clays. These relationships can be explained on the basis of particle arrangement patterns: face-to-face arrangements dominated the Ca-clays and edge-to-edge and edge-to-face arrangements dominated the Na-clays. The amount of iron extractable in dithionite-citrate-bicarbonate (DCB) correlated positively with the relative number of plates per tactoid and with the major dimensions of the particles in the Ca-clay suspensions. This correlation suggests that DCB-extractable iron affects the tactoid dimensions of Ca-clays from vertisols and, therefore, may also affect structural properties of vertisols.

Resistant starch (RS) has received increased attention due to its potential health benefits. This study was aimed to investigate the effects of dietary corn RS on immunological characteristics of broilers. A total of 320 broiler chicks were randomly allocated to five dietary treatments: normal corn–soyabean (NC) diet group, corn starch diet group, 4 %, 8 % and 12 % RS diet groups. This trial lasted for 42 d. The relative weights of spleen, thymus and bursa, the concentrations of nitric oxide (NO) and IL-4 in plasma at 21 d of age, as well as the activities of total nitric oxide synthase (TNOS) and inducible nitric oxide synthase (iNOS) in plasma at 21 and 42 d of age showed positive linear responses (P < 0·05) to the increasing dietary RS level. Meanwhile, compared with the birds from the NC group at 21 d of age, birds fed 4 % RS, 8 % RS and 12 % RS diets exhibited higher (P < 0·05) relative weight of bursa and concentrations of NO and interferon-γ in plasma. Birds fed 4 % RS and 8 % RS diets showed higher (P < 0·05) number of IgA-producing cells in the jejunum. While compared with birds from the NC group at 42 d of age, birds fed 12 % RS diet showed higher (P < 0·05) relative weight of spleen and activities of TNOS and iNOS in plasma. These findings suggested that dietary corn RS supplementation can improve immune function in broilers.

Methionine is a limiting amino acid (AA) in fish diets, particularly in those containing high levels of plant protein (PP), and is key in the immune system. Accordingly, outcome on the fish immune mechanisms of methionine-deficient and methionine-supplemented diets within the context of 0 % fishmeal formulation, after a short and prolonged feeding period, was studied in European seabass (Dicentrarchus labrax). For this, seabass juveniles were fed a (i) fishmeal-free diet, meeting AA requirements, but deficient in methionine (MET0·65); (ii) as control, the MET0·65 supplemented with l-methionine at 0·22 % of feed weight (CTRL); (iii) two diets, identical to MET0·65 but supplemented at 0·63 and 0·88 % of feed weight of l-methionine (MET1·25 and MET1·5, respectively); and (iv) a fishmeal-based diet (FM), as positive control. After 2 and 12 weeks of feeding, blood and plasma were sampled for leucocyte counting and humoral parameter assays and head-kidney collected for gene expression. After 2 weeks of feeding, a fishmeal-free diet supplemented with methionine led to changes in the expression of methionine- and leucocyte-related genes. A methionine immune-enhancer role was more evident after 12 weeks with an increased neutrophil percentage and a decreased expression of apoptotic genes, possibly indicating an enhancement of fish immunity by methionine dietary supplementation. Furthermore, even though CTRL and FM present similar methionine content, CTRL presented a reduced expression of several immune-related genes indicating that in a practical PP-based diet scenario, the requirement level of methionine for an optimal immune status could be higher.

The objective of this study was to determine the protective effect of glutamate (Glu) in Cu-induced oxidative injury in fish intestine in vivo and enterocytes in vitro. The results indicated that exposure to 6 mg/l Cu for 72 h induced the production of reactive oxygen species, thereby increasing protein oxidation and lipid peroxidation in enterocytes of grass carp in vitro. Cells exposed to Cu alone resulted in a significant increase in lactate dehydrogenase release, which is accompanied by depletions of antioxidants, including total superoxide dismutase (T-SOD), glutathione S-transferase (GST), glutathione reductase (GR), anti-superoxide anion (ASA), anti-hydroxy radical (AHR) activities and GSH content. Pre-treatment with Glu remarkably prevented the toxic effects of Cu on the T-SOD, GST, GR, AHR, and ASA activities and GSH content in enterocytes. However, Cu induced an adaptive increase in the activities of catalase and glutathione peroxidase (GPx). Glu supplementation further increased GPx activity in enterocytes. Interestingly, the experiment in vivo showed that Glu pre-supplementation significantly elevated SOD, GPx, GST, GR, ASA and AHR activities, as well as GSH content. Further results showed that pre-treatment with Glu could alleviate Cu-induced oxidative injury by elevating antioxidant enzyme activities through regulating the expression of NF-E2-related nuclear factor 2 (Nrf2) mRNA. Together, these results indicated that Glu could attenuate Cu-induced cellular oxidative damage in fish intestine, likely mediated through Nrf2 signalling pathways regulating mRNA expressions of antioxidant enzyme genes and synthesis of GSH.

A new approach to spectroscopy of laser induced proton beams using radiochromic film (RCF) is presented. This approach allows primary standards of absorbed dose-to-water as used in radiotherapy to be transferred to the calibration of GafChromic HD-810 and EBT in a 29 MeV proton beam from the Birmingham cyclotron. These films were then irradiated in a common stack configuration using the TARANIS Nd:Glass multi-terawatt laser at Queens University Belfast, which can accelerate protons to 10–12 MeV, and a depth-dose curve was measured from a collimated beam. Previous work characterizing the relative effectiveness (RE) of GafChromic film as a function of energy was implemented into Monte Carlo depth-dose curves using FLUKA. A Bragg peak (BP) “library” for proton energies 0–15 MeV was generated, both with and without the RE function. These depth-response curves were iteratively summed in a FORTRAN routine to solve for the measured RCF depth-dose using a simple direct search algorithm. By comparing resultant spectra with both BP libraries, it was found that the effect of including the RE function accounted for an increase in the total number of protons by about 50%. To account for the energy loss due to a 20 µm aluminum filter in front of the film stack, FLUKA was used to create a matrix containing the energy loss transformations for each individual energy bin. Multiplication by the pseudo-inverse of this matrix resulted in “up-shifting” protons to higher energies. Applying this correction to two laser shots gave further increases in the total number of protons, N of 31% and 56%. Failure to consider the relative response of RCF to lower proton energies and neglecting energy losses in a stack filter foil can potentially lead to significant underestimates of the total number of protons in RCF spectroscopy of the low energy protons produced by laser ablation of thin targets.

Variability among samples analysed using the same ELISA protocol generates ambiguity in deciding which assay best quantifies the protein concentration. In this study, we propose a standardization method, called I-STOD (Improved STandardization method for Optical Density), for the transformation of OD values on different plates into relative concentrations of the antibody levels being assessed. We derived an equation relating OD values of different test samples to antibody levels according to the multi-stage reaction dynamics of the indirect-ELISA. Using serum samples from a Schistosomiasis japonica endemic area, we evaluated the fitness of the I-STOD model to experimental data of a standard reference serum in comparison with 5 other models. Calibration curves fitted by the I-STOD method judged to be superior, based on adjusted R2 (adjusted R2>0·99 on 22 out of 26 plates) values. The CV (coefficient of variation) value of the results between multi-well plates and the number of plates with OD values beyond the control range in Shewhart charts also demonstrate that the I-STOD method is a powerful tool which can greatly improve the comparability of results on different multi-well ELISA plates. We conclude that a standardization method is certainly necessary for antibody levels detected in order to properly illustrate clinical differences.

We use the photopigment template of Baylor et al. (1987) to define the set of Rayleigh matches that would be satisfied by a photopigment having a given wavelength of peak sensitivity (λmax) and a given optical density (OD). For an observer with two photopigments in the region of the Rayleigh primaries, the observer's unique match is defined by the intersection of the sets of matches that satisfy the individual pigments. The use of a template allows us to illustrate the general behavior of Rayleigh matches as the absorption spectra of the underlying spectra are altered. In a plot of the Y setting against the red–green ratio (R), both an increase in λmax and an increase in optical density lead to an anticlockwise rotation of the locus of the matches satisfied by a given pigment. Since both these factors affect the match, it is not possible to reverse the analysis and define uniquely the photopigments corresponding to a specific Rayleigh match. However, a way to constrain the set of candidate photopigments would be to determine the trajectory of the change of match as the effective optical density is altered (by, say, bleaching or field size).
